The key molecule in this interaction is ubiquitin that serves to activate the interaction between the protein and the proteasome. This is being done using several different small molecules that can link the ubiquitin recruiting protein E3 ligase and the disease causing protein. Most popular are PROTACS, that are heterobifunctional small molecules, that links disease causing proteins to an E3 ligase. Another emerging technology is molecular glues that are monofunctional small molecules that is able to link E3 ligases and proteins by binding one, and modifying their affinity towards the other, thereby activating the ubiquitin proteasome pathway.

Protein degradation cycle. E3 ligase adds ubiquitin to the protein of interest. When the protein is polyubiquinated with at least 4 ubiquitins, the protein of interest is degraded in the protesome an cleaved into peptides and free ubiquitin.
